井伊於10月29日、近江國犬上郡的彥根城（現滋賀縣）出生。
彥根中最大的湖是琵琶湖。
並成為井伊直中（第十三任當主）的第十四個兒子

因為井伊是庶民所生，出生次序又後，所以有很大的一段時間都在郊外生活。
並自號埋木舍，比喻自己為深埋地底、無法開花的木材。

1846年，井伊成為了第15代井伊氏掃部頭家當主。
1858年，井伊成為了第13任江戶幕府大老。其地位相當於宰相，萬人之上，一人之下。

他在彥根藩時代進行了藩政改革，被認為是一代名君。
曾主張開國、私自同美國簽署日美修好通商條約及剷除異己。
而其原因各歷史學家抱有不同意見。

期後孝明天皇對於直弼所作所為十分不滿，偷偷詔令水戶藩剷除無視武家秩序的直弼。在1860年3月3日被水戶藩浪人在江戶城櫻田門附近暗殺，史稱「櫻田門外之變」。

現在彥根城內和橫濱市的掃部山公園（日語：掃部山公園）為了表彰他實行開國政策的功勳，而建有他的銅像，但是對於實行開國政策的利弊也有不同的看法。
從以前到現在都有很多不同的聲音評論井伊。
